White adipose tissue WAT is an endocrine organ highly integrated in homeostasis and capable of establishing ways of communicating and influencing multiple metabolic processes. Brown adipose tissue promotes energy expenditure by incorporating the uncoupling protein 1 UCP1 , also known as thermogenin, which decouples cellular respiration and heat production, in the mitochondrial membranes. Recent data suggest the presence of a thermogenic cell formation from white adipocytes beige or brite cells with a potential role in preventing obesity and metabolic syndrome. Irisin, a thermogenic adipomyokine produced by FNDC5 cleavage is involved in the browning of adipose tissue. While animal studies are congruent with regard to the relationship between physical exertion and irisin release, the results from human studies are less than clear. Therefore, this review focuses on recent advances in our understanding of muscle and adipose tissue thermogenesis.

The rising prevalence of chronic diseases such as type 2 diabetes and cardiovascular diseases owing to fat mass excess has been described. Irisin was described as an exercise-induced myokine. The main beneficial function attributable to irisin is the change of subcutaneous and visceral adipose tissue into brown adipose tissue, with a consequential increase in thermogenesis. Irisin has also been described as a hormone that may have a key role in glucose homeostasis. The way the association of type 2 diabetes with obesity occurs is not fully understood. In recent years, the possible pathways through which irisin could interact with other organs such as the brain or bone have been described. The present paper intends to review the new findings and possible new directions in irisin research. Obesity is a health problem present in developed and developing countries [ 1 ]. Insulin resistance has been considered the link between obesity and chronic degenerative diseases [ 2 ]. The rising prevalence of chronic diseases such as type 2 diabetes and cardiovascular diseases owing to fat mass excess has been described [ 3 ]. Over the past decade, most of the research efforts related to metabolic diseases have been focused on adipose tissue and its role in chronic inflammation [ 4 ].

In , a transmembrane protein—now known as FNDC5—was discovered and shown to be expressed in skeletal muscle, heart, and brain. It was virtually ignored for 10 years, until a study in proposed that, in response to exercise, the ectodomain of skeletal muscle FNDC5 was cleaved, traveled to white adipose tissue, and induced browning. The wasted energy of this browning raised the possibility that this myokine, named irisin, might mediate some beneficial effects of exercise. Since then, more than papers have been published exploring the roles of irisin. A major interest has been on adipose tissue and metabolism, following up the major proposal from Many studies correlating plasma irisin levels with physiological conditions have been questioned for using flawed assays for irisin concentration.

An Author Correction to this article was published on 07 October Here, we show that irisin, the cleaved and circulating form of the exercise-induced membrane protein FNDC5, is sufficient to confer the benefits of exercise on cognitive function. Diminished pattern separation in F5KO mice can be rescued by delivering irisin directly into the dentate gyrus, suggesting that irisin is the active moiety. In F5KO mice, adult-born neurons in the dentate gyrus are morphologically, transcriptionally and functionally abnormal. Importantly, elevation of circulating irisin levels by peripheral delivery of irisin via adeno-associated viral overexpression in the liver results in enrichment of central irisin and is sufficient to improve both the cognitive deficit and neuropathology in AD mouse models. Irisin is a crucial regulator of the cognitive benefits of exercise and is a potential therapeutic agent for treating cognitive disorders including AD.

The vitality of these primary hippocampal nerve cells from diabetic rats was markedly decreased when BDNF levels were low but improved following irisin treatment. On the other hand, subcutaneous adiposity appears to be benign in terms of the incidence and severity of complications 6. Albrecht et al 16 measured the apparent MW of 2 well-characterized r-irisins in Western blots. There are only a few exceptions of cellular response at assumed physiological concentrations. Myokine irisin-induced protection against oxidative stress in vitro. PTEN loss in the Myf5 lineage redistributes body fat and reveals subsets of white adipocytes that arise from Myf5 precursors. Ursolic Acid-induced elevation of serum irisin augments muscle strength during resistance training in men. Irisin and Brain Physical exercise has been associated with the reduction of physical and cognitive complications related to central nervous system disorders [ 63 ]. In mice, irisin is released from skeletal muscle during exercise acts directly on bone by increasing cortical bone mineral density, bone perimeter and polar moment of inertia. According to Huh et al. First, the initial study suggested rather high amounts of irisin in circulation of mice and humans even detectable by Western blots 3. BMC Cancer. Non his-tagged irisin was recovered in the flow through fraction from the cobalt column.
